چکیده

A cultivation-independent approach was used to identify potentially nitrogen-fixing endophytes in seven sweet potato varieties collected in Uganda and Kenya. Nitrogenase reductase genes (nifH) were amplified by PCR, and amplicons were cloned in Escherichia coli. Clones were grouped by restriction fragment length polymorphism analysis, and representative nifH genes were sequenced. The resulting sequences had high homologies to nitrogenase reductases from alpha-, beta-, and gamma-Proteobacteria and low G+C Gram positives, however, about 50% of the sequences derived from rhizobia. Several highly similar or even identical nitrogenase reductase sequences clustering with different bacterial genera and species, including Sinorhizobium meliloti, Rhizobium sp. NGR234, Rhizobium etli, Klebsiella pneumoniae, and Paenibacillus odorifer, could be detected in different plants grown in distinct geographic locations. This suggests that these bacterial species preferentially colonize African sweet potato as endophytes and that the diazotrophic, endophytic microflora is determined only to a low degree by the plant genotype or the soil microflora.

Complete Genome Sequence of Kosakonia sacchari Strain BO-1, an Endophytic Diazotroph Isolated from a Sweet Potato

The complete genome sequence of the endophytic diazotroph Kosakonia sacchari, isolated from a sweet potato, was analyzed. The 4,902,106-bp genome with 53.7% G+C content comprises 4,638 open reading frames, including nif genes, 84 tRNAs, and seven complete rRNAs in a circular chromosome.

Diversity of the Bacterial Microbiome in the Roots of Four Saccharum Species: S. spontaneum, S. robustum, S. barberi, and S. officinarum

Endophytic bacteria are nearly ubiquitously present in the internal tissues of plants, and some endophytes can promote plant growth. In this study, we sampled the roots of four ancestral species of sugarcane (two genotypes per species) and two sugarcane cultivars, and used 16S rRNA and nifH gene sequencing to characterize the root endophytic bacterial communities and diazotroph diversity. A tot...

Strong shift in the diazotrophic endophytic bacterial community inhabiting rice (Oryza sativa) plants after flooding.

Flooding impacts soil microbial communities, but its effect on endophytic communities has rarely been explored. This work addresses the effect of flooding on the abundance and diversity of endophytic diazotrophic communities on rice plants established in a greenhouse experiment. The nifH gene was significantly more abundant in roots after flooding, whereas the nifH gene copy numbers in leaves w...

Phylogenetic analysis of nifH gene sequences from nitrogen-fixing endophytic bacteria associated with the roots of three rice varieties

Abstract The community structure of nitrogen-fixing endophytic bacteria associated with roots of three varieties, Oryza sativa cv. Sprice and cv. Koshihikari and line NERICA 5 (an interspecific hybrid between O. sativa and O. glaberrima) was analyzed using culture-independent molecular techniques. A segment of nifH gene was amplified from crude rice root DNA, cloned to construct nifH library an...

Genome-wide assessment of population structure and genetic diversity and development of a core germplasm set for sweet potato based on specific length amplified fragment (SLAF) sequencing

Sweet potato, Ipomoea batatas (L.) Lam., is an important food crop that is cultivated worldwide. However, no genome-wide assessment of the genetic diversity of sweet potato has been reported to date. In the present study, the population structure and genetic diversity of 197 sweet potato accessions most of which were from China were assessed using 62,363 SNPs. A model-based structure analysis d...
